Gene that causes a form of deafness discovered
ScienceDaily (Sep. 30, 2012) ? Researchers at the University of Cincinnati and Cincinnati Children's Hospital Medical Center have found a new genetic mutation responsible for deafness and hearing loss associated with Usher syndrome type 1.
These findings, published in the Sept. 30 advance online edition of the journal Nature Genetics, could help researchers develop new therapeutic targets for those at risk for this syndrome.
Partners in the study included the National Institute on Deafness and other Communication Disorders (NIDCD), Baylor College of Medicine and the University of Kentucky.
Usher syndrome is a genetic defect that causes deafness, night-blindness and a loss of peripheral vision through the progressive degeneration of the retina.
"In this study, researchers were able to pinpoint the gene which caused deafness in Usher syndrome type 1 as well as deafness that is not associated with the syndrome through the genetic analysis of 57 humans from Pakistan and Turkey," says Zubair Ahmed, PhD, assistant professor of ophthalmology who conducts research at Cincinnati Children's and is the lead investigator on this study.
Ahmed says that a protein, called CIB2, which binds to calcium within a cell, is associated with deafness in Usher syndrome type 1 and non-syndromic hearing loss.
"To date, mutations affecting CIB2 are the most common and prevalent genetic cause of non-syndromic hearing loss in Pakistan," he says. "However, we have also found another mutation of the protein that contributes to deafness in Turkish populations.
"In animal models, CIB2 is found in the mechanosensory stereocilia of the inner ear -- hair cells, which respond to fluid motion and allow hearing and balance, and in retinal photoreceptor cells, which convert light into electrical signals in the eye, making it possible to see," says Saima Riazuddin, PhD, assistant professor in UC's department of otolaryngology who conducts research at Cincinnati Children's and is co-lead investigator on the study.
Researchers found that CIB2 staining is often brighter at shorter row stereocilia tips than the neighboring stereocilia of a longer row, where it may be involved in calcium signaling that regulates mechano-electrical transduction, a process by which the ear converts mechanical energy -- or energy of motion -- into a form of energy that the brain can recognize as sound.
"With this knowledge, we are one step closer to understanding the mechanism of mechano-electrical transduction and possibly finding a genetic target to prevent non-syndromic deafness as well as that associated with Usher syndrome type 1," Ahmed says.
Other researchers involved in the study include Thomas Friedman, PhD, and Inna Belyantseva, MD, PhD, from the NIDCD; Suzanne Leal, PhD, and her team at Baylor; and Gregory Frolenkov, PhD, and his team at the University of Kentucky.
This study was funded by the NIDCD, the National Science Foundation and the Research to Prevent Blindness Foundation.

Idaho woman charged with stick-figure threat
BOISE, Idaho (AP) ? A woman who was about to be released from an Idaho prison will stay behind bars a while longer because federal prosecutors say she mailed a threatening stick figure drawing to a relative.
Linda Joyce Lakes was serving time at the Pocatello Women's Correctional Center on probation violations stemming from a 2007 grand theft sentence, and she was scheduled to be released last Friday. But earlier this month, the U.S. attorney's office charged her with mailing threatening communications, a felony.
Prosecutors say that in 2010, Lakes mailed a drawing that apparently depicted a battered stick figure alongside threatening phrases such as "No tears," ''No hiding," and "No more you."
Kyle Wright, an FBI agent in Pocatello who investigated the case, described the drawing in a court document.
"One stick figure appeared to be lying down with his face smashed. The baseball bat was lying nearby with damage that appeared to be consistent with its use on the first stick figure. The second stick figure appeared to be walking away with a smile," Wright wrote.
Wright also said in his affidavit that Lakes sent a letter with the drawing that indicated her prison sentence was coming to an end and that the recipient should be prepared for his final moments.
The FBI agent also said that Lakes told the FBI her drawing symbolized her hate for a man who had abused her in the past, and that one day she wanted to beat and possibly kill him.
Lakes was released from the state prison and taken into federal custody. She waived her preliminary hearing on the charge and has not yet entered a plea.
Lisa Tolle, a paralegal with Federal Defender Services of Idaho in Pocatello, said her office had just been assigned the case and attorneys had not yet fully reviewed it.

Mercer Island upends Sammamish | Prep football
In a game that included nine touchdowns and four fumble recoveries, the Mercer Island football team advanced to a 3-1 record on Friday, Sept. 21, beating Sammamish at home 63-0.
Senior quarterback Alec Bean earned the first start of his career with Mercer Island, leading off the game by assisting for a touchdown, the first of many points of the evening. Peter Mahony scored just two minutes later after the Islanders defense kept Sammamish from advancing far. Zack Bucklin hit the mark with 5:48 left in the first off a pass from quarterback Brian Higgins, pushing the score to 20-0.
After the first of what would become four fumble recoveries for the Islanders with 4:43 left in the first, Bucklin scored his second of the night. Running back Devlin Conway, a junior, followed suit with 2:58 left in the first and it ended with the Islanders leading 35-0.
Just 16 seconds into the second quarter, Mercer Island?s Mahony made a 67-yard touchdown run, upping the score to 42-0 with 11:44 left to play in the half. The second Totem fumble of the night game with 2:49 left in the half, and the Islanders began their quest on the 45 yard line. The Islanders attempted a field goal as time ran out in the half, but the ball was tipped and didn?t reach the posts. The score was 42-0 at halftime.
During the break the school honored it?s five new Mercer Island High School Hall of Fame inductees. The five included: Chris Bailey, Duke Biggers, Trevor Fulp, Kelsey Ingram and Greg Mahony.
Back on the field after halftime and the clock running because of the 42-point rule, Mercer Island hit the mark again with 6:13 left in the third when Risley Lesko ran it in and was tackled in the corner. The score was 49-0.
Lesko scored his second of the night with 1:25 left in the third, this time on a longer run for a 56-0 lead for Mercer Island. Mercer Island didn?t score again until Bean, back in the game at quarterback, ran the ball into the zone to give the team its final points of the night.
Next Friday, Sept. 28, the Islanders will once again be at home, hosting Interlake in a 7 p.m. matchup.

SANTA CLARA, Calif. (AP) - Chris Wondolowski scored two goals in the second half, including the tying goal in stoppage time, to lift the San Jose Earthquakes to a 2-2 draw with the Portland Timbers on Wednesday night.
Wondolowski, who entered the game in the 58th minute, scored his 20th and 21st goals - extending his franchise single-season record, career-high and league-leading total - to help San Jose (17-6-6) improve to 11-0-5 in its last 16 matches at Buck Shaw Stadium. The Earthquakes also remained three points ahead of Sporting Kansas City in the Supporters' Shield race for the most points in the standings.
Portland (7-14-8), which got two goals from Danny Mwanga, squandered an opportunity for its first road win of the season. Instead, the Timbers remained winless in a 16th straight road match and are on pace to be just the fifth MLS club to go a full season without a road victory.
San Jose salvaged the critical point after an attempted flick clearance by Portland defender Hanyer Mosquera floated to the back post, where an open Wondolowski headed it home.
San Jose brought in Wondolowski and Alan Gordon - initially rested in the midst of a three-games-in-eight-days stretch - looking for late-match magic, and they each had a hand in the Earthquakes' first goal.
Gordon flicked on a long lead pass from Victor Bernardez in the 73rd minute, and Steven Lenhart got a sliding touch on it to free Wondolowski, who converted with his left foot from 16 yards. It was Wondolowski's first goal in the run of play since July 14.
Mwanga used his strength to create his own goal-scoring chance in the 45th minute, staying upright despite a hard challenge from Bernardez. Bernardez's attempt to knock the ball from Mwanga fed straight to Rodney Wallace in the middle of a stretched-out San Jose defense. Wallace slid a through ball right back to Mwanga, who took one dribble to avoid converging defenders and charging goalkeeper Jon Busch and found the empty net.
Mwanga, acquired from Philadelphia in June, made it 2-0 with a crackling 25-yard blast across the face of goal to the upper corner of the far post in the 62nd minute. The play was set up by a 40-yard run through the midfield by Franck Songo'o. Earthquakes midfielder Sam Cronin futilely gave chase, eventually coming up short on a sliding tackle, and Songo'o easily swiveled around Bernardez before feeding Mwanga, who had plenty of time to line up his shot.

Toronto Mayor Rob Ford has no credibility and can't be believed when he says he did no wrong in connection to allegations he violated conflict of interest rules, says the lawyer who has cross-examined Ford. Ford was questioned for four hours on Wednesday by Clayton Ruby about his refusal to repay $3,150 in donations to his football foundation ? something Janet Leiper, the city's integrity commissioner, had first asked him to do in 2010, and then repeated that request on several occasions. The court will decide if Ford's participation in a debate on Feb. 7 over the issue and in a subsequent council vote that let him off the hook for the money put the mayor in a conflict of interest as defined by the Municipal Conflict of Interest Act (MCIA). Ruby told a Toronto courtroom on Thursday that he doesn't believe Ford made an honest error of judgment when he spoke and voted on the issue. Ruby said Ford, who was first elected to council in 2000, and then as mayor in 2010, has no credibility on the matter. The mayor's "peculiar definition of conflict" doesn't mesh with his extensive experience on council, Ruby said. Ford asserted in court on Wednesday that his interpretation of a conflict of interest, as defined by the act, only occurs if a council member speaks or acts on a matter in which both the city and that member stand to benefit. Students create low-cost biosensor to detect contaminated water in developing nations
ScienceDaily (Sep. 6, 2012) ? Diarrheal disease is the second-leading cause of death in children under five years old -- killing as many as 1.5 million children worldwide every year. These startling statistics from the World Health Organization (2009) point to the reason why a group of undergraduate students from Arizona State University is working to develop a low-cost biosensor -- a simple device that would detect contaminated drinking water.
An interdisciplinary team of nine students is participating in the 2012 International Genetically Engineered Machine (iGEM) competition -- a global event that challenges students to design and build simple biological systems made from standard, interchangeable parts.
The ASU team started its research during the summer to prepare for the synthetic biology competition. Its goal is to create a user-friendly, DNA-based biosensor that can detect major pathogens. The low-cost device would be used in the field rather than in a laboratory.
"We are developing a biosensor that will detect pathogenic bacteria, such as Shigella, Salmonella and E. coli, that cause diarrhea," said Ryan Muller, an undergraduate student in ASU's School of Life Sciences and an iGEM team leader. "Ideally, you would use our biosensor to check different water supplies in third world-countries to determine whether the water is safe to drink."
The team is working on two biosensor designs.
"The first one targets DNA," explained Nisarg Patel, a molecular biosciences and biotechnology major in School of Life Sciences, as well as a political science major. "Since each type of pathogen has different DNA, we want to create complementary sequences -- sequences that match a specific DNA. We will take bacterial samples from the water, pull out the DNA and check whether it complements our DNA probe. If it does, it will produce a color response and then we'll know that the water is contaminated."
Made for portability, Patel said the second design tests the membranes of bacteria. When using the device to test water, if certain proteins attach to a bacterial membrane, the sample will turn blue -- indicating the water is contaminated with a pathogen and would not be safe to drink.
"The advantage of this design over previous designs in the field lies in the cheap production of probes and the enzymatic chain reaction," said Abhinav Markus, a biomedical engineering student in ASU's Ira A Fulton Schools of Engineering. "Samples can be tested in the field with minimal cost and high sensitivity."
When the ASU iGEM team first met this summer, Madeline Sands, an anthropology major in the university's School of Human Evolution and Social Change, pitched the idea to build a low-cost biosensor. Sands previously traveled to Guatemala as part of an ASU field experience. There, she conducted community health research under the direction of Jonathan Maupin, a medical anthropologist. Sands realized that contaminated water presents a serious health problem for developing countries.
"With constant earthquakes, landslides and rains in Guatemala, it can often be difficult to determine if a water source is contaminated," said Sands. "My time there made it clear that having a way to detect contaminated water could lead to a further reduction in the incidence and morbidity of diarrhea."
In October, the team will present its device during the iGEM regional competition at Stanford University. If successful, they will move on to the global competition in November at Massachusetts Institute of Technology.
Other ASU iGEM team members include: Rohit Rajan, Ethan Ward, Hyder Hussain, Amanda Ispas and Ellen Qin. Kylie Standage-Beier, a biological sciences major and previous iGEM team member, serves as an advisor.

Bubble Tea Tapioca Pearls May Cause Cancer, Study Says
Bad news for fans of the colorful novelty drink called tapioca tea, or boba tea: The sugary specialty beverage, generally milk-based and filled with chewy balls of tapioca, may also include cancer-causing chemicals known aspolychlorinated biphenyls or PCBs, the Daily News reports.
German researchers from the University Hospital Aachen have reportedly found traces of the carcinogenic chemical in tapioca ball samples. The tapioca was taken from an unnamed chain in northwest Germany and originated in Taiwan, according to the Daily News.
"[What we found] includes in particular styrene, acetophenone, and brominated substances that shouldn't be in food at all," scientist Manfred M?ller, of the Institute of Hygiene and Environmental Medicine at the University Hospital Aachen, told German newspaper The Local, notes the AFP.
According to the EPA, PCBs belong to a broad family of man-made organic chemicals manufactured between 1929 and 1979. The chemicals still exist in the environment despite their U.S. manufacturing ban. Ranging in toxicity, PCBs have been shown to cause cancer, as well as a variety of other adverse health effects on the immune system, reproductive system, nervous system and endocrine system, the government site continues.
Bubble tea has gained widespread popularity in recent years across Europe, and Germany in particular, after already converting young people across North America. German McDonald's have even begun selling the dessert beverage as part of its recently revamped McCafe menu, the Independent reports.
The cancer concerns were compounded by another public health warning, released earlier in August by the country's German Federal Institute for Risk Assessment. German authorities warned that the beverage's hallmark gummy balls, may pose a choking risk.
"Especially with children aged up to four years, there is a risk of foreign objects accidentally entering the lungs," said Dr. Andreas Hensel in a press release on the Institute's website. "And that is precisely what can happen when the bubbles are sucked up through a straw."

Both the living trust and joint tenancy avoids probate. ?A trust is more flexible than joint tenancy and easily divides the property among the list of beneficiaries. ?Joint tenancy is less flexible, cannot include post-death trusts for tax planning, and works best when two or three people are involved.
The person creating the trust generally retains control and flexibility and has not made a taxable gift to anyone. ?The trustor can generally change the terms of the trust and change the beneficiary at any time without the consent of the beneficiary, because the beneficiary has no vested right to the property.
A person who creates a joint tenancy makes a present transfer of property. ?This may result in a gift and may be subjected to taxes. ?The new owner may possesses the right of partition and can make it difficult to sell/transfer the property. ?Therefore, if you decide to create a joint tenancy, it must be with an individual whom you trust. ?Furthermore, a joint tenant?s creditors may reach his or her interest in the property.
